چکیده
SUMMARY The unicellular cyanobacterium Synechococcus PCC 7942 has been used as a model organism for studies of prokaryotic circadian rhythms, carbon-concentrating mechanisms, response to a variety of nutrient and environmental stresses, and cell division. This paper presents the results of the first proteomic exploratory study of Synechococcus PCC 7942. The proteome was analyzed using two-dimensional gel electrophoresis followed by MALDI-TOF mass spectroscopy, and database searching. Of 140 analyzed protein spots, 110 were successfully identified as 62 different proteins, many of which occurred as multiple spots on the gel. The identified proteins were organized into 18 different functional categories reflecting the major metabolic and cellular processes occurring in the cyanobacterial cells in the exponential growth phase. Among the identified proteins, 14 previously unknown or considered to be hypothetical are here shown to be true gene products in Synechococcus sp. PCC 7942, and may be helpful for annotation of the newly sequenced genome.

Promoters of the phycocyanin gene clusters of the cyanobacterium Synechococcus sp. strain PCC 7942.
The cyanobacterium Synechococcus sp. strain PCC 7942 has duplicated phycocyanin subunit gene clusters cpcB1A1 and cpcB2A2, which are identical to each other and to those of Synechococcus sp. strain PCC 6301 (Anacystis nidulans). Nucleotide sequences of the 428 and 286 bases of the 5' non-coding regions of the cpcB1A1 and cpcB2A2 clusters, respectively, of strain PCC 7942 were identical to those...
متن کاملDifferent strategies of metabolic regulation in cyanobacteria: from transcriptional to biochemical control
Cyanobacteria Synechococcus sp. PCC 7942 and Synechocystis sp. PCC 6803 show similar changes in the metabolic response to changed CO2 conditions but exhibit significant differences at the transcriptomic level. This study employs a systems biology approach to investigate the difference in metabolic regulation of Synechococcus sp. PCC 7942 and Synechocystis sp. PCC 6803. Presented multi-level kin...
متن کاملPhosphorylation of the PII protein (glnB gene product) in the cyanobacterium Synechococcus sp. strain PCC 7942: analysis of in vitro kinase activity.
The PII protein in the cyanobacterium Synechococcus sp. strain PCC 7942 signals the cellular state of nitrogen assimilation relative to CO2 fixation by being phosphorylated at a seryl residue. In this study, we first determined the location of the phosphorylated seryl residue within the PII amino acid sequence. The phosphorylation site exhibits an RXS motif, a recognition sequence characteristi...
متن کاملSequencing and Modification of the Gene Encoding the 42-Kilodalton Protein in the Cytoplasmic Membrane of Synechococcus PCC 7942.
A 42-kilodalton cytoplasmic membrane protein is synthesized when high CO(2)-grown cells of Synechococcus PCC 7942 (Anacystis nidulans R2) are exposed to low CO(2). The structural gene for this protein (cmpA) has been cloned and sequenced and shown to encode a 450 amino acid polypeptide with a molecular mass of 49 kilodalton. A deletion mutant lacking the 42-kilodalton protein was obtained by tr...
متن کاملComparative Analysis Of Heavy-Metal Ion Sensing Mechanisms With Transcription Factors, Smtbs, From Freshwater Synechococcus Sp. PCC 7942, And Marine Synechococcus Sp. PCC 7002: Evolutionary And Structural Aspects
Metal-responsive transcription factors play a crucial role in the metal ion homeostasis with both cellular metabolism and environmental metal availability. In present work, we studied structural and evolutionary features of a transcription repressor, SmtB from freshwater Synechococcus sp. PCC 7942 and its homologs from other cyanobacteria in detail. We mined putative SmtB-like zinc sensors from...
